A goal of biology is to predict how mutations combine to alter phenotypes, fitness and disease. It is often assumed that mutations combine additively or with interactions that can be predicted. I will describe our work showing that this assumption is incorrect. Mutations combine differently at different expression levels. Sometimes predicting how mutations combine would be impossible even with perfect measurements of the mutational effects on a trait and the mechanistic understanding of the genetic system.
